My protocol for making 9M urea in a final 200uL volume is listed below:1)2mL tube with round bottom (easier for dissolving urea with vortex), and weigh 108 mg urea2)Add 70uL 1% MC (final concentration is 0.35%)3)Add 20uL deionized water4)Vortex to dissolve (if urea is crystal not powder, you can use sonication to make it dissolve faster. Do not heat the solution over 30℃, that make urea degrade), the final volume is 180uL5)Add pharmalyte 8uL (final concentration is 4%)6)Add sample 12uL (If sample concentration is too low, you need to concentrate it first)
