摘要:

A best method of analyzing anthocyanidins of Lycium ruthenicum Murr. was studied by using high performance liquid chromatography(HPLC) and study on structure of anthocyanin in Lycium ruthenicum Murr. by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS). The HPLC-MS was used to analysis kind and content of anthocyanidins from Lycium ruthenicum Murr..The anthocyanidins was extracted by ultrasonic assistance extraction using powder of Lycium ruthenicum Murr. dissolved The conditions of samples hydrolyzed and detection by HPLC was studied using the chromatogram quality as index. The results showed that anthocyanidin of Lycium ruthenicum Murr. was hydrolyzed in hydrolysate solution comprised of alcohol solu-tion, distilled water and hydrochloric acid solution according to the volum ratio of 3:2:1. The concentration of HCl in hydrolysate solution was 3.0mol/L. It was hydrolyzed in boiling water for 1 h. The flow speed of liquid phase was 0.7 mL/min. There were good results of HPLC detection according to the conditions that had been se-lected. The anthocyanins in Lycium ruthenicum Murr. had been detected by the HPLC. The result showed that the content of delphinidin, cyanidin, petunidin, and malvidin was 2.32,15.44,35.18,2.63 μg/mL in Lycium ruthenicum Murr. respectively.The HPLC-MS method was used to analyze the components of anthocyanidins in Lycium ruthenicum Murr.. The optimum conditions for the determination of anthocyanin were determined by ex-periment. The results showed that the anthocyanin content of Lycium ruthenicum Murr. was determined and the results were obtained and detected 14 kinds of anthocyanin components.%研究高效液相色谱(high performance liquid chromatography,HPLC)检测黑果枸杞中花青素含量的最佳方法,采用高效液相色谱-串联质谱(high performance liquid chromatography-tandem mass spectrometry,HPLC-MS)分析黑枸杞中的花色苷结构.以超声波辅助提取获得的花青素粉末作为样品,对影响检测结果的样品水解时间,液相流速等因素进行研究,并采用高效液相色谱-串联质谱检测花青素.确定水解液比例为乙醇:水:盐酸=3:2:1(体积比),水解液中HCl浓度为3.0 mol/L,水解时间1 h,流速为0.7 mL/min时,液相检测效果较好.结果表明,黑果枸杞中飞燕草素,矢车菊素,矮牵牛素和锦葵素的浓度分别为2.32,15.44,35.18,2.63 μg/mL.通过试验确定高效液相色谱法的最佳检测条件,确定黑果枸杞花青素的含量;并对黑果枸杞花青素进行一定的结构分析,得到较好的结果,并检测出14种花色苷成分.
